Normal Perfusion of Isolated Rat Hearts and Its Influence on Interfibrillar and Sub-sarcolemmal Mitochondria Activities

چکیده
Obvious lack of interfibrillar mitochondria (IFM) sub population in isolated mitochondria attributed to limitations in isolation procedure. In this manuscript, we compared the functional activities of mitochondrial sub-populations namely, IFM and sub-sarcolemmal (SSM) mitochondria obtained from isolated rat heart subjected to different perfusion time namely (minutes) 0.5,5,10,25,60 and 120. Prolonged perfusion of isolated rat heart reduced oxidative phosphorylation capacity in both IFM and SSM, but was distinct among the subpopulations. However, mRNA expression level of ND4, CYT B and ATP 6 and resazurin activity were similar in both IFM and SSM. Even though overall function of myocardium is unchanged, mitochondrial subpopulations were distinct in electron transport chain activities, emphasizes the requirement to assess mitochondrial function as distinct subpopulation rather than whole entity.

The Effect of Verapamil Administred before the Reperfusion Insult in Isolated Preconditioned Rat Heart on the Microsomal ATPase and Mitochondrial Enzyme Activities
Background: Calcium overload and free radical mediated damage in the mitochondria is the most important pathological changes associated with myocardial ischemic-reperfusion injury. The verapamil post-treatment has been previously reported to prevent reperfusion-induced myocardial injury but functional recovery may be delayed due to the drug's inherent direct myocardial depression effect. In the...

Objective(s): To determine the effects of triiodothyronine (T3) intracoronary perfusion in isolated hearts and short-term administration in rats on the left ventricular (LV) phosphorylation patterns of Akt and ERK1/2. Materials and Methods: Cardiodynamic and hemodynamic parameters were evaluated in Langendorff–perfused hearts. Left ventr...
متن کاملThe effect of asafoetida essential oil on myocardial ischemic-reperfusion injury in isolated rat hearts
Objective: Previous studies reported that asafetida from Ferula assa-foetida Linn. species and its essential oil (AEO) have antioxidant effects. In the present study, the effect of AEO was evaluated on ischemic-reperfusion injury in isolated rat hearts. Materials and Methods: Forty-eight male Wistar rats were divided into 6 groups: 1) control group, 2) vehicle group, 3-5) AEO groups and, 6) car...
متن کاملA Defect in Oxidative Phosphorylation in the Cardiac Interfibrillar Mitochondria*
To determine whether or not an oxidative defect exists in hearts of cardiomyopathic (BIO 14.6) hamsters, cardiac tissue and subcellular organelles were studied. Using homogenates of hearts, the sarcolemmal marker, 5’-nucleotidase, was decreased, whereas the lysosomal enzymes, acid phosphatase and N-acetylglucosaminidase, were increased in the cardiomyopathic hearts compared to controls. The mit...
متن کاملDefects in sarcolemmal Ca2+ transport in hearts due to induction of calcium paradox.
Na+-Ca2+ exchange and Ca2+-pump activities were studied in sarcolemmal vesicles isolated from rat hearts subjected to "calcium paradox" on perfusion with Ca2+-free medium followed by reperfusion with medium containing 1.25 mM Ca2+. Perfusion of hearts with Ca2+-free medium for 5 minutes did not affect the Na+-dependent Ca2+ uptake, ATP-dependent Ca2+ uptake, or Ca2+-stimulated ATPase activities...
